Steve,

The lower, bigger nut is a lock nut. Turn it in a clock-wise direction, so it goes closer to the bottom of the boat. This will unlock the "cap" (stuffing box) containing the packing. Then turn the cap in the same direction. Probably as tight as you can get it. I would then check to see if the leak stops befor re-tightening the lock nut. If you can not stop the leak then you may need to repack the stuffing box (cap).
